Dlx Family
The Dlx or Dll gene family is named after the single Drosophila representative distalless (Dll). Amphioxus also has a single member (AmphiDll; Holland et al. 1996). In human, there are six Dlx genes arranged as three pairs, each linked to a Hox gene cluster: DLX1 and DLX2 are linked to HOXD, DLX3 and DLX4 are linked to HOXB, DLX5 and DLX6 are linked to HOXA. Phylogenetic analysis reveals that these descended from an ancestral linked pair of genes (thus DLX1, 4 and 6 form a robust clade, while DLX2, 5 and 3 form a clade in some analyses; Stock et al. 1996, Holland et al. 2007). It is currently unclear if the common ancestor of Drosophila and vertebrates had one or two Dlx genes. Dlx genes have been widely studied because of their roles in limb development (Panganiban et al. 1997), although they are also involved in many other developmental processes.
